Here is the Formulation and Standard Operating Procedure (SOP) for BarIno™ FusaTrinity™, a dual-action agarwood inoculant combining chemical and microbial agents:
Product Name: BarIno™ FusaTrinity™
“Dual-Action Resin Induction – Microbial Power Meets Oxidative Priming”
Formulation & Preparation SOP
Product Composition (per 1 liter batch):
Component | Function | Amount |
---|---|---|
Fusarium oxysporum Culture | Biotic inducer of resin production | 100 mL (live culture or spore suspension) |
Manganese Dioxide (MnO₂) | Oxidative stress agent (mild) | 2.0 g |
Magnesium Chloride (MgCl₂) | Stress priming + nutrient support | 3.0 g |
Brown Sugar | Microbial nutrient/carbon source | 10.0 g |
Sterile Distilled Water | Base solvent | To make up 1.0 L |
Optional: Tween 20 (0.1%) | Surfactant for better tissue penetration | 1 mL (optional) |
Culture Preparation (if not using pre-supplied):
- Fusarium oxysporum Isolation & Culturing
- Use PDA (Potato Dextrose Agar) medium to culture Fusarium oxysporum from a verified resin-inducing strain.
- Incubate at 25–28°C for 5–7 days.
- Harvest spores or mycelial suspension by washing culture with sterile distilled water and filtering through cheesecloth.
- Spore Count Standardization (Optional but ideal):
- Adjust suspension to ~1×10⁶ spores/mL using a hemocytometer or serial dilution method.
Final Inoculant Preparation (per 1 L):
- Dissolve MnO₂, MgCl₂, and brown sugar in 500 mL sterile distilled water under agitation.
- Add live Fusarium oxysporum suspension (100 mL) into the mixture.
- Top up to 1 liter with sterile distilled water.
- Mix gently to avoid damaging fungal spores.
- Add Tween 20 if using, and mix again.
Storage & Shelf Life:
- Fresh Use Recommended: Within 48–72 hours at room temp (25–30°C).
- If refrigerated (4°C), can last up to 7 days, though microbial activity may decrease over time.
- Do not freeze.
Application Guidelines:
Parameter | Details |
---|---|
Target Trees | Aquilaria malaccensis, A. crassna, ≥ 3 years old, minimum 6–8 cm DBH |
Application Method | Drill holes 5–8 mm wide, 2–3 cm deep into the trunk at 20–30 cm intervals |
Inoculant Volume | 3–5 mL per hole (depending on tree size and spacing) |
Post-Application | Seal holes with wax/clay or breathable cotton plug |
Frequency | One application every 6–12 months depending on tree health and objectives |
Synergistic Effect:
- MnO₂ acts as a mild oxidant, priming the plant’s defense pathways and improving Fusarium colonization.
- MgCl₂ + Sugar nourish both the tree and the inoculated fungus.
- Results in a faster, broader, and longer-lasting resin induction compared to single-mode inoculants.
