Formulation & Preparation SOP


Product Composition (per 1 liter batch):

Culture Preparation (if not using pre-supplied):

  1. Fusarium oxysporum Isolation & Culturing
    • Use PDA (Potato Dextrose Agar) medium to culture Fusarium oxysporum from a verified resin-inducing strain.
    • Incubate at 25–28°C for 5–7 days.
    • Harvest spores or mycelial suspension by washing culture with sterile distilled water and filtering through cheesecloth.
  2. Spore Count Standardization (Optional but ideal):
    • Adjust suspension to ~1×10⁶ spores/mL using a hemocytometer or serial dilution method.

Final Inoculant Preparation (per 1 L):

  1. Dissolve MnO₂, MgCl₂, and brown sugar in 500 mL sterile distilled water under agitation.
  2. Add live Fusarium oxysporum suspension (100 mL) into the mixture.
  3. Top up to 1 liter with sterile distilled water.
  4. Mix gently to avoid damaging fungal spores.
  5. Add Tween 20 if using, and mix again.

Storage & Shelf Life:

  • Fresh Use Recommended: Within 48–72 hours at room temp (25–30°C).
  • If refrigerated (4°C), can last up to 7 days, though microbial activity may decrease over time.
  • Do not freeze.
