Standard Operating Procedure (SOP) for the Sodium Chloride (NaCl) inoculant, used as an abiotic stressor to induce agarwood resin formation in Aquilaria spp.

SOP: NaCl-Based Agarwood Inoculant

Product Name: BarIno™ SaltStrike™
Tagline: “Stress it smart. Scent it strong.”
Purpose: To prepare a low-cost, moderately effective abiotic stress inoculant for inducing resin in agarwood trees via NaCl osmotic and ionic stress.

1. Materials Needed

MaterialSpecification
Sodium Chloride (NaCl)Non-iodized, food or lab grade (≥98%)
Distilled or clean waterNeutral pH (6.5–7.5)
Brown sugar (optional)Organic or refined, powder form
Clean mixing containerPlastic or glass (non-metallic)
Stirring rod or hand mixerNon-reactive (e.g., glass or plastic)
Measuring toolsDigital scale, graduated cylinder
PPEGloves, goggles, mask

2. Formulation Procedure

Standard Formula:

  • NaCl concentration: 3–5% w/v
(i.e., 30–50 g NaCl per liter of water)
  • Optional brown sugar: 2% w/v
(i.e., 20 g per liter) for microbial synergy

Preparation Steps:

  1. Weigh Ingredients:
    • NaCl: 30 g (mild) to 50 g (moderate stress) per 1 L
    • Brown sugar: 20 g (optional)
  2. Dissolve Solutes:
    • Add ~900 mL distilled water into the container.
    • Slowly add NaCl while stirring to dissolve fully.
    • If using brown sugar, add and stir until clear.
  3. Adjust Final Volume:
    • Add more water to make exactly 1 liter of solution.
  4. Filter (optional):
    • For precision or sterile conditions, pass solution through cheesecloth or fine filter.

3. Storage & Shelf Life
