Standard Operating Procedure (SOP) for formulating and applying Salicylic Acid (SA) Inoculantas an agarwood resin inducer in Aquilaria spp.

SOP: Salicylic Acid–Based Agarwood Inoculant

Product Name: BarIno™ SignalResin™
Tagline: “Signal the Tree. Trigger the Resin.”
Purpose: To simulate a biotic stress signal and trigger systemic acquired resistance (SAR) and phenolic resin biosynthesis in agarwood trees.

1. Materials Required

MaterialSpecification
Salicylic Acid (SA)Analytical or food-grade (≥99% purity)
Ethanol or Isopropanol95% (for dissolving SA)
Distilled WaterpH 6.5–7.0
Brown Sugar (optional)2% w/v to enhance microbial growth if co-inoculated
pH meter or pH stripsFor checking final solution acidity
Beaker & Stirring RodNon-reactive glass or plastic
Digital ScaleFor accurate weighing
PPEGloves, goggles, mask

2. Inoculant Formulation

Target Concentration: 0.2% w/v SA Solution

IngredientQuantity (per 1 Liter)
Salicylic Acid2 grams
Ethanol (95%)20–30 mL
Distilled WaterUp to 1000 mL
(Optional) Brown Sugar20 g (for 2% w/v)

Preparation Steps:

  1. Dissolve SA in Alcohol:
    • In a small beaker, dissolve 2 g of salicylic acid in 20–30 mL of ethanol (SA is poorly water-soluble without alcohol).
    • Stir until fully dissolved.
  2. Add Water:
    • Pour the solution into a clean container and dilute with distilled water to make up to 1 liter.
    • Mix thoroughly.
  3. (Optional) Add Sugar:
    • Add 20 g of brown sugar to the solution if using in combination with microbial inoculants.
  4. Check pH:
    • Final pH should be between 5.5–6.5. Adjust if needed using dilute NaOH or citric acid.
